Airyscan tool
Quality and status indicates the current status of the detector alignment.
Unknown (Waiting): No alignment done or possible. Make sure the Airyscan detector is activated and
the gain is set high enough to detect emission light. A continuous scan allows for the self alignment to
be performed.
Bad (Waiting/Adjusting): Automatic alignment is in progress.
Waiting: The system is waiting for position analysis feedback.
Adjusting: Adjustement is performed. During the alignment the system toggles between these two
processes.
Good (Waiting/Adjusting): Automatic aligment is in progress. Waiting: System is waiting for position
analysis feedback; Adjusting: Adjustement is performed. During the aligment the system toggles between
these two processes.
For image acquisition the status should be indicated as Good (Waiting).
Adjustments will only be performed during continuous scans (if activated) or during delay times of several
seconds between imaging cycles of a long term time series acquisition (if activated).
To activate the automatic self adjustment of the Airyscan, make sure at least one of the automatic
alignment modes is activated. The system will self adjust during continuous scans or slow time series
acquisition.
Note, that this display and adjustment is not available in VP and CO mode. To get optimal
adjustment in VP and CO mode, it is recommended to switch to SR mode from time to time and
run a continuous scan.
The detector view of the Airyscan tab shows the intensity distribution of the detection elements. This also
allows a judgement whether the system is adjusted correctly. The brightest element should be in the
center with a drop of the signal towards the periphery.
In case the detector has not been used for longer periods of time in SR mode or not at all, a continuous
scan should be run for alignment before image acquisition is started in SR mode.
The sliders in the Servo position panel (Show all activated) are used to manually adjust the detector in
X (X (µrad)) and Y (Y (µrad)).
A position (XY) is already stored and can be defined again as default (Store Current Pos). This position
can be reset in case the automatic alignment fails because the detector is too far off. Click Move To
Stored Pos and start continuous scanning again.

Maintain – Airyscan
The Airyscan tool provides options for automatic
alignment of the Airyscan detector.
As a superresolution detector the alignment within
the detection beam path is critical and it is
recommended to have both alignment options,
Adjust in continuous scans and Adjust in slow
time series activated at all times.
The function will not be activated
automatically upon starting the system if
it has been deselected before shutting
down the system. When starting the
software, a message will remind you of
activating the adjustment options.
