ZEISS
Maximal in the drop down list next to the Power status. The maximal available laser power changes to a
higher percentage value and the value is now indicated in red as well as the higher tube current. This
operation mode reduces the life time of the laser and should only be used for short periods of
time.
Laser life time
The absolute maximum power varies with the laser and declines over time as the laser is in use. A given
tube current will over time produce less and less laser output power. Maximal power should only be set
for short periods of time. Once the laser is no longer able to produce 100% output power with the
maximal tube current available, the percentage of power and the tube current will also show up in red.
With a tool tip you will be reminded, that the laser might reach the end of its lifetime. It can still be
operated but its lifetime will decline even faster as it will be operated at high tube current to achieve the
100 or near 100 % of power.

Experiment Designer tool
Changing acquisition parameters always effects acquisition parameters of the active acquisition block
even if the Experiment Designer tool is not open and/or not enabled.
When Enable Multi Block Experiment is checked, all functions within the tool become accessible.

Laser dynamics
The regulation of the all visible laser lines (458 nm
up to 633 nm) is achieved via AOTF. The amount
of power is proporational to the electric power
generating the acousto-optic wave in the AOTF
cristal. The AOFT cristal can deal with up to 8 laser
lines in parallel. However, if the power settings for
the individual lasers differ by at least an order of
magnitude, inerferences can occur between the
lines and the laser power can vary during imaging
when applied. In this case a warning shows up in
the Channels tool window (Fig. 92) and the higher
power setting for the AOTF will be reduced.

Setup Manager – Experiment
Designer
Experiment Designer combines different multi-
dimensional acquisition parameters with each
parameter setting assigned to one imaging block in
one total experiment described as Multi Block
Experiment. Click the Experiment Designer tab
to open or close the tool window (Fig. 93).
There is always one blue Block (Active Acquisition
Block)
present
with
parameters assigned to it.
