Processing – Lightsheet Processing

The Lightsheet Processing module allows one to process images that were acquired with the
Lightsheet Z.1 using Dual Side illumination and/or with the Multiview tool tab.
Files need to be saved prior to any Lightsheet Processing task. If this is not the case the software will ask
to save the data before proceeding.
All images and registration files resulting from Lightsheet Processing can be found in a Result
folder that is created in the folder of the original file, that was used for processing.

Dual Side Fusion

This processing task can be used for the following
datasets acquired with Dual Side illumination:
− Image (Snap function)
− Time Series
− Z-Stacks
− Combinations of Time Series and Z-Stacks
− Multiview experiments as well in
combination with Time Series
When a dataset has been acquired using Dual
Side illumination, the dataset will have the
illumination dimension saved in its metadata and
the Dimension slider for Illumination is visible in
the Image Control area. Both illumination sides
are handled as single frames within the image.
Dual Side Fusion provides three main options for
merging
both
images
illumination sides into a single frame.
Choose the dataset you wish to perform the task
by mark either option (Fig. 515).
Discrete Fourier Transform Fusion:
The fusion is performed based upon the published method ("Multiple imaging axis microscopy improves
resolution for thick-sample applications", Jim Swoger, Jan Huisken, and Ernst H. K. Stelzer, OPTICS
LETTERS / Vol. 28, No. 18 / September 15, 2003).
Fine structures and details are more visible after the fusion which might be worsened when using Mean
Fusion. However, this process can create noticeable artifacts in the resulting image.
