Here, the Autofocus every n Timepoint check box is checked and a value of 3 entered in the
associated spin box.
A focusing event will be executed before the first acquisition and after the completion of three
acquisition cycles. In this example, control of activity intervals is only linked to the acquisition of the time
series.
The performance of the Fluorescence, Reflection and Autofocus Map autofocus modes can be readily
tested with a defocused sample.
After pressing the
button, the system will be guided to the imaging plane.
An image acquisition will be triggered automatically as soon as the system arrives at this Z-position.
Next, the system will travel back to the Z-level it started from when
was clicked.
Test Autofocus is only available if Show all is active.
Testing applies the currently selected autofocus mode and performs the test at the current XY position.
With the modes Reflection and Autofocus Map, the value for Offset will be taken into consideration.
Status information with LSM-based autofocus modes
The first focus action with one of the LSM-based modes is performed as a two-step procedure including a
coarse focusing (XZ line scan) followed by a fine focusing step (XY frame scan).
