CHAPTER 1 - SYSTEM OPERATION
Left Tool Area and Hardware Control Tools
Smart Setup displays its proposals side-by-side for
optimal comparison. The proposal selected for imaging is
highlighted. Key aspects of each proposal are displayed
as a bar chart representation.
Such bar charts contain information on the expected
emission signal (relative amount of detectable signal with
the
contribution of cross talk) and speed (acquisition speed
as percentage of acquisition speed in simultaneous multi-
channel recording).
The prediction of the emission signal considers both, the
efficiency of fluorochrome excitation and also the
detectable portion of the emission.
Smart Setup displays a bar chart representation of
emission signal, cross talk and speed with each proposal.
The color-coding specified for the stains is used for
displaying the bar charts (Fig. 74).
Smart Setup displays the track configuration along with
normalized emission spectra of the chosen dyes (as
specified in the experiment configuration). Filled spaces
of the spectral curves illustrate the portion of the
emission detectable in a given proposal (Fig. 75). The
illustration also displays the order of track acquisitions
upon sequential multi-color recording. Track acquisitions
using long wavelength laser lines are performed first.
Pressing the
proposal as well as the motif. If a motif other than
Current is selected, ZEN applies the proposal, the
scanning parameters of the motif and automatically
scans an image. Detector settings are automatically
adjusted by pressing the
