Processing – Experimental PSF
 standard
Tool accepts Z-Stacks. See full description of tool for details.
This function allows determining the point-spread
function (PSF) of sub-resolution beads and using
this experimentally determined PSF as input for
Structured Illumination processing.
Select the Experimental PSF tab to select the
Experimental PSF Processing tool (Fig. 467).
Activate the image (a 3D stack of sub resolution
beads) you want to process in the image container
and press the Select button for Input Image to
load the image. Then expand the Method
Parameters tool window to have access to setting
parameters (Fig. 468).
You can load a 3D stack of a widefield
image or a non-processed raw data SIM
image. LSM image formats are not
accepted to determine the Experimental
PSF.
You have the possibility to automaticaly compute
the Experimental PSF by choosing Automatic from
Mode drop down menu (Fig. 469).
If you want to process manually and set quality
parameters, select Manual (Fig. 470).
Pressing the Default button will load Standard
values. Data will only be reused, if the result image
is selected. Than pressing the Reuse button will
load the values of the selected image. The options
Symmetrical and Damp Edge are not included in
the Reuse function.
When the Symmetrical check box is ticked, a
radial symmetrization of the PSF will be performed.
That means that a perfectly round PSF is
calculated, even out of an oval PSF.
When the Damp edge check box is ticked, the
edge of the PSF will be smoothed (with a smooth
transition to zero at the edge).
