Find the optimal excitation wavelength.
This procedure uses the emission maximum to determine the optimum excita-
tion wavelength, and is similar to step 1.

Insert the sample into the sample compartment, and close the sample
compartment's cover.
Click the Run button
The scan starts.
With the spectrum on the screen, note the greatest intensity.
If the signal exceeds 2 × 10
saturating, so close the slits more. If there is no obvious peak, increase
the excitation wavelength, starting, and ending by 25 nm, and retry a
scan.
With an acceptable emission peak, record its wavelength.
This is the emission maximum. Otherwise, repeat steps i through l until
an obvious emission peak appears.
In the toolbar, click the Experiment Menu button
The Fluorescence Main Exper-
iment Menu appears:
Choose the Spectra button.
The Experiment Type window
appears.
Choose Excitation.
Click the Next >> button.
The Experiment Setup window appears:
