In the ROI Inspector dialog box:

In the Exposure Time field, enter 2048.

Determine if your ROI image is acceptable (the

figures to the right show an unsaturated image
and an oversaturated image). Wells in an
acceptable image:
• Must be as bright as possible without
oversaturating. (When you generate the ROI
calibration on

• Can contain some, but do not have to
contain any, red pixels, which represent
saturation.

If your ROI image is acceptable, skip to

If your ROI image is oversaturated, decrease the
Exposure Time by half, then click
. Repeat until you obtain an
acceptable ROI image.
