Discrete Sample Scan

To scan a single sample dissolved in a buffer, follow
these instructions.
1
Locate one 1/4-28 coupler and one male Luer
end-fitting from the accessory package. Make
the following connections:
• Attach the coupler to the detector outlet
• Connect the Luer end-fitting to the coupler
• Attach a syringe to the Luer end-fitting

Disconnect the detector inlet tubing from the
column. Immerse the end of the inlet tubing
into a pure buffer solution.

Draw the buffer solution through the inlet
tubing into the syringe.

From the first display for scan mode, press the
Zero soft key to perform a zero scan and to
reset the baseline of the trace at the monitor
wavelength to zero. The mobile phase spectrum
is automatically subtracted from the compound
spectrum during the scan.

Immerse the inlet tubing into the sample of
interest (in the same buffer).

Draw the sample solution through the inlet
tubing into the syringe. Be sure that the cell is
overfilled on both ends to eliminate bubbles
from the flow cell.

Press Scan to begin the sample scan.
