4.1 Introduction

4.2 Method Selection

The ACCQPrep HP125 is unique in the Preparative HPLC
market. There are several methods of operation possible that can
be selected to fit your needs.
Methods contain all of the information used during a separation
of a single sample, such as gradient profile, detection and peak
cutting parameters, and fraction size. Repetitive sample injection
is programmed as needed and doesn't require creation of a sep-
arate file.
One of the unique features of the ACCQPrep is that a defined
method file is not required for operation. A default or existing
method can be loaded and user modified for the current sepa-
ration without the need for saving the newly modified method.
In addition, any of the method parameters can be modified
during the separation. None of these changes will affect the
original method unless you choose to save the changes to the
original method. All method and injection parameters used for
starting the separation or modified during the separation are
stored with the chromatographic data so no information is lost.
In addition, the method and injection parameters used during a
separation can be extracted from the data file and saved as a
named method for future reuse.
Several options can be used to select a method:
• Use a pre-programmed column that has several methods
associated with it. Once the column is selected, you can
simply select one of the associated methods. This is
especially convenient if you typically only use a few
predefined methods.
• Use a method file that has been saved before by going to

· Use the file dialogue to open the run file from a
previous separation and select the E
on the lower portion of the screen.
If the method selected is adequate, simply press the P
to continue. If the method is not exactly as needed, the method
can be edited. Edit the common parameters of any method either
by adjusting the points on the graph, changing major parameters
on the main screen, or by selecting the M
edited, the method can be used directly for the immediate sepa-
ration(s) or saved with a name for future reuse.
