• Vacuum filtration kit; for example, WAT085113, which includes a filter holder, flask, vacuum
pump, and filters
• Additional Membrane filters, 0.1 μm or smaller, PVDF
To filter neutral or basic mobile phase:
1.
Rinse all wetted surfaces of the vacuum filtration system with MS-grade isopropanol and
allow the surfaces to dry.
2.
Install the vacuum filtration system according to the instructions in the kit.

Vacuum filter the mobile phase through a 0.1 μm or smaller PVDF membrane.
Result:
down through the membrane into the flask.
4.
Rinse the bottle to which you will transfer the mobile phase with MS-grade isopropanol and
allow the bottle to dry.
5.
Transfer the mobile phase to the clean bottle.
D.2.4.4 Preparing new mobile phase every two days
To limit the time for bacterial growth, prepare neutral and basic pH mobile phases at least every
two days. Switch the fresh mobile phase for the old one, and then prime the pump to remove the
old mobile phase in the system. Ensure that you use bottles rinsed with MS-grade water.
Tip:
Do not allow solvent lines in neutral or basic mobile phases to rest on the bottom of the
bottle.
