Image quality control
The fluorescence signal in the reference channel is measured to determine the number of valid
partitions in a well. Differences in the signal intensities between partitions are normalized and the
fluorescence signals in the target channels are corrected accordingly.
If the fluorescence signal is saturated in too many partitions of a well in a target channel, a
warning message will be shown to the customer. Furthermore, saturated signals are marked with
in the Results overview. Saturated signals lower the signal-to-noise ratio and can lead to
improper analysis results, e.g. the crosstalk correction algorithm might be affected. The
recommendation is to re-image the plate with 30% less exposure time or gain value in the
respective channel.
In case more than one imaging step has been done, the imaging step where the saturation occurred
is marked in yellow and the warning message is shown when moving the mouse over the camera
icon.
