To create a program with the Program Wizard
1. Open the Program Wizard. On the File menu, select New, and then select Program
File.
2. The wizard guides you through program creation. On each wizard page, make the
desired settings or accept the default values. For additional information about a page,
click Help. The page to be used for the fluorescence detector settings is named FLD
Options.
First, select whether you want to record one or several emission channels. Select the
parameters for the channel or channels in the table. For information about parameter
settings, see page 77. The wizard page changes depending on whether you select one or
several channels for recording data.
If you are recording a single channel, enter the expected (typical) minimum peak width at
half height of the peak in the Peak Width box. Chromeleon calculates an appropriate
data collection rate and response time on the basis of these two parameters. Click Apply
to accept these values (recommended). Then select a lamp mode in the Lamp Mode box
(→ page 88).
In addition, you can define the flow cell temperature (→ page 88) and the allowed
deviation.
