Viewing the Specimen
12
View the specimen.
To observe another part of the sample, rotate the stage
knobs to find the position to observe. If the sample is not
in focus, rotate the focus knobs to adjust the focus.
To observe under a different magnification, repeat steps
6 and later with another objective.
Diascopic image in fluorescence
observation
For fluorescence observations, press the
microscope's dia-illumination ON/OFF switch to
"OFF" to make the diascopic image disappear.
Bright ambient lights will make it difficult to view the
image. Nikon recommends keeping the room dark
during fluorescence observations.
NI-FA FL/DIC Analyzer Slider/Dummy
Slider
In the epi-fluorescence microscopy, the FL/DIC
analyzer slider inserted in the epi-fluorescence cube
turret is not used. When the analyzer slider is
inserted in the epi-fluorescence cube turret, pull it out
to the first click position to remove the analyzer from
the optical path. When a dummy slider instead of the
analyzer slider is inserted, do not fully insert it. Pull it
out one step. If it is fully inserted, the periphery of
field of view may be dark.
