Running Samples
This chapter describes the typical procedures of running examples on NovoCyte
Quanteon instrument using human peripheral blood lymphocyte subsets analysis
as an example.

Overview

This test uses the Agilent CD3 FITC/CD8 PE/CD45 PerCP/CD4 APC Reagent and
the Agilent CD3 FITC/CD16+CD56 PE/CD45 PerCP/CD19 APC Reagent to stain
human peripheral blood lymphocyte subpopulations. The kit contains two sets of
four-color antibodies, which are CD3 FITC/CD8 PE/CD45 PerCP/CD4 APC and CD3
FITC/CD16 +CD56 PE/CD45 PerCP/CD19 APC respectively. For a detailed
description of the reagents and sample preparation process, please refer to the
reagent technical data sheet.

This example will create a specimen named human peripheral blood, which
consists of two tubes of human peripheral blood samples prepared and stained
respectively by the Agilent CD3 FITC/CD8 PE/CD45 PerCP/CD4 APC Reagent and
the Agilent CD3 FITC/CD16+CD56 PE/CD45 PerCP/CD19 APC Reagent. The test
needs to use 488 nm, 561nm and 637 nm lasers, and collect signals of the Height
and Area parameters on FSC, SSC, and the fluorescent channels including B525
(FITC), Y586 (PE), B667 (PerCP), and R667 (APC). Each sample will be collected at
a Slow sample flow rate until the events reach 5,000 in the Lymphocyte gate.
