Running Samples
If you select the Unstained sample in the Compensation Channel, calculation of the
NOTE
compensation matrix will be based on the unstained sample, instead of the
negative population in the single-stained sample.
The Compensation Channels shown in the New Auto Compensation window will
NOTE
be all the available channels based on the model of the NovoCyte Quanteon flow
cytometer. The NovoExpress software automatically obtains such available
channel information from the hardware. Correctly select the Compensation
Channels to ensure the correct automatic compensation results. You can also
change the settings by right-clicking the Compensation Specimen in the
Experiment Manager (Figure 51) and select Auto Compensation Setup ....

Run Samples for Automatic Fluorescence Compensation
The default setting for acquiring automatic compensation samples are:
In order to ensure normal data acquisition and correct automatic fluorescence
NOTE
compensation, it is recommended to prepare the compensation sample
concentration to be 1 x 10^6 ~ 2 x 10^6 /mL.
Most samples can be properly collected with the default settings. However, users
NOTE
can also change the settings accordingly.
