Introduction
NovoFlow sheath fluid: For hydrodynamically focusing the sample stream. Use
1X NovoFlow directly as sheath fluid or dilute 100X NovoFlow one hundred
folds with 0.2 μm filtered, deionized water.
NovoRinse solution: For rinsing and removing protein deposits, adherent cell
debris, etc. in the fluidics system.
NovoClean solution: For cleaning the fluidics system.
When the volume of NovoFlow sheath fluid, NovoRinse solution, or NovoClean
solution is less than the set threshold (NovoFlow 0.9 L, NovoRinse 150 mL,
NovoClean 150 mL), the NovoExpress software will prompt a warning message to
remind the user to add the appropriate reagents. Similarly, when the waste volume
exceeds a set threshold (2.8 L), the software will give a warning to remind the user
to empty the waste container. The colors of the LED status indicator on the front
of the NovoCyte Fluidics Station II also give information on the fluid level and are
summarized in the following table.

Green
Reagent volume is above the designated threshold and is sufficient
to run experiments (NovoFlow > 0.9 L; NovoRinse > 150 mL;
NovoClean > 150 mL), and the waste container is not full (< 2.8 L).
Orange
Reagent volume is below the threshold but is still sufficient to finish
the current running experiment. (0.7 L < NovoFlow < 0.9 L; 100 mL <
NovoRinse < 150 mL; 100 mL < NovoClean < 150 mL), or the waste
container is close to full (2.8 L < waste < 3 L). Please add more
reagents to the corresponding container or empty the waste
container.
Red
Reagent volume is too low to run experiments (NovoFlow < 0.7 L;
NovoRinse < 100 mL; NovoClean < 100 mL), or the waste container
is full (> 3 L). Any operation of the instrument is prohibited until
there are enough reagents added or the waste container has been
emptied.
