• Liquid—Select this option when you will be injecting
a liquid sample into the injection port.
• Solid—Select this option when you will be using a
solid sample cartridge.
• Direct —This option is useful when a sample is direct
loaded on the column media. When this option is
selected, the Companion XL skips the column
equilibration and proceeds with the purification run.
5. Click the Play button.
PeakTrak will list the minimum solvent and tube quantities
necessary for the run. At this time you can also specify which
tube in the rack to begin with. Ensure that the requirements
are met, set the first tube, and click OK.
If the Sample Type was set for Liquid or Solid, the
Companion XL then equilibrates the column with the initial
conditions.
After equilibrating, the system will pause if you selected the
Liquid option for the Sample Type. This pause waits for you to
inject the sample. Click OK to continue the run.
If you selected the Solid option for the Sample Type, the
system proceeds with the purification after equilibrating the
column if a cartridge is present. If a cartridge is not present,
the system will pause for the user to load the cartridge.

8.6 Operation During the Purification Run

After injecting the sample, the Companion XL delivers the
first 3 mL of liquid into the first tube, then advances to the
next tube. This clears the line of eluant held in the drop
former tube from a previous run.
You may allow Companion XL to proceed with the run while
monitoring the progress on the PeakTrak Main window. If
desired, gradient parameters may be modified during the run.

As the run progresses, the absorbance trace is drawn on the
Gradient Plot area. The separation or purification continues
until the end of the run defined by the run length setting,
expressed in Column Volumes.
