Chase solvent clears the Liquid Injection accessory tubing and
forces the liquid compound mixture onto the column. Without the
chase solvent, some of the sample will remain in the tubing and
will not be separated. This small amount of sample may also
cross-contaminate the next purification run.
The chase solvent should be strong enough to prevent the
sample from precipitating (or crashing), but not so strong as to
adversely affect the separation of compounds. Often, the initial
liquid phase solvent is a practical chase solvent.
4. Inject a chase solvent:
a. Connect a syringe loaded with a chase solvent to the
Liquid Injection accessory's luer fitting.
b. Open the valve by turning the handle so it is in line with
the tubing (Figure 8-7).
c. Inject at least 10 mL of the chase solvent.
d. Close the valve by turning the handle so that it is
perpendicular to the tubing. Remove the syringe.
5. Click the OK button to continue the run.
