7.2.9 Preparing the Sparger

Procedure

CAUTION!
Danger of loss of property from a dirty inlet fil-
ter due to leaking of culture liquid!
Culture liquid can leak out of the vessel via the
sparger through the inlet filter due to pressure dur-
ing sterilisation if the silicone hose is not clamped
off entirely. The inlet filter will get defective and will
need replacement in that case.
Therefore:
– Ensure the inlet air silicone hose is well
clamped off.
The sparger must be equipped with a hose and inlet filter before
sterilisation.
Fit the inlet filter not too tightly, as the connection will serve as a
pressure relief point in case of overpressure
To prepare the sparger, proceed as follows:
1.
Cut a piece of thick-walled silicone hose.
2.
Fit the silicone hose onto the sparger in the vessel.
3.
Secure the silicone hose with a cable tie.
