7.3 Sterilisation in the Autoclave

7.3.1 Sterilisation Methods

The vessel must be custom-designed sterilised in an autoclave be-
fore starting the fermentation/cultivation process.
WARNING!
Microbial danger!
An incorrect sterilisation procedure may lead to a
microbial risk to the user when fermenting and
could also result in unwanted and unexpected
changes to the process.
Therefore:
– Carry out the sterilisation process thoroughly
– Sterilise for a minimum of 30 minutes at a rec-
ommended minimum of 121 °C
– Respect custom-designed sterilisation condi-
tions.
– Strictly follow local safety procedures.
CAUTION!
Risk of damage to the pump heads!
Sterilising the pump heads at too high tempera-
tures or together with hoses filled with corrective
reagent will lead to damage to the pump heads.
Therefore:
– Never sterilise the pump heads at temperatures
above 125 °C
– Fill corrective reagent hoses with water for steri-
lisation.
Sterilisation is to be carried out custom-designed. There are differ-
ent possible sterilisation methods. The following strategies are
commonly used:
