To start a program
Program for sample analysis
1. Create a sample list (sequence). A sequence must include the program and a method
for evaluating the sample data (for example, for peak identification, area
determination, and amount determination).
2. Assign the program and method to each sample on the list.
3. Add the sequence to the batch and start the batch.

5.3 Shutting Down the Fraction Collector

To shut down the fraction collector, complete the following steps:
1. Flush flow paths as required and as outlined in section 5.4.
2. Shut down the HPLC system to which the fraction collector is connected as is required
and as instructed via the associated documentation and outlined procedures therein.
3. Turn off the AFC-3000 power switch.

5.4 Flushing the Rinse Station and Eluent Flow Path

Generally, you can operate the AFC-3000 without flushing the rinse station. Under normal
circumstances, you can simply drain the rinse station prior to shutting down the fraction
collector. However, you should flush the rinse station and flow path under two
circumstances:
• during initial startup of the AFC-3000, after installation
• following the use of strong bases, acids, or organic solvents as may be present in the eluent
Flushing the rinse station during initial startup of the AFC-3000 removes any
contaminants. Flushing the rinse station after using strong rinse/eluent agents/solvents
prevents degradation and failure of the flow path components. If additives such as buffers
are used as a part of the mobile phase, flush the system with several volumes of a suitable
solvent before shutting the AFC-3000 down. This will prevent buildup of precipitates
inside the flow path.
To flush the rinse station and flow path, flush the entire HPLC system as outlined in the
system's operating instructions.
