Sampling Tips
Background Measurement
Sample Measurements
Thermo Scientific
For optimal sampling with the instrument, follow these guidelines:
• Leave the instrument powered on continuously.
• For optimum instrument stability, the sampling arm should remain closed except when
cleaning it or loading a sample. Keeping the arm closed also allows background
measurements to occur automatically.
• Clean the sample windows thoroughly before you run the
• To apply a sample for measurement, use a clean dropper bottle, pipette or glass rod to
place a small volume of sample in the sample cup. A micropipette that dispenses
approximately 70 μl works well. The cup should be about two thirds full. (Do not overfill
the sample cup.)
• Before you start a measurement, gently close the sampling arm and then wait a few
seconds for the sample to settle. Measurements can be made only when the arm is down
(an error message appears if the arm is up when the measurement starts). Do not touch
the sampling arm until the measurement has completed.

The workflows display a message when a background collection is needed. Make sure the
sampling arm is closed and follow any workflow instructions to ensure a good quality
background is acquired.
An FT-IR background measurement is needed to correct for variation in the instrument signal
over time. Information from the background is used to separate absorptions that are due to
the instrument laboratory environment from absorptions that are due to the sample.
A background measurements is required at predefined intervals during continuous use. We
recommend that the time between background collections should not exceed 2 hours.
