Introduction

The Avanti JXN series centrifuge specification for temperature control is ± 2°C. The following
procedure is provided for those cases in which temperature control within ± 1°C is required.
In this procedure, you will measure the temperature of your sample after equilibrating the rotor
temperature for a minimum of 1 hour. Based on the test results, you can then adjust the centrifuge
temperature setting to bring your sample as close as possible to the required temperature.
Calibrating Temperature

For runs at other than ambient temperature, precool or prewarm the rotor and test samples
to the required temperature.
The length of precooling/prewarming time depends on how different the rotor starting
temperature is from the required run temperature.

Install the rotor with adapters, if applicable, and tubes or bottles filled with sample buffer or
water (if above 2°C).

Set the speed to 2000 RPM, the time to
temperature.
a. Start the run.

After 30 minutes, set the required run speed, and allow the system to run for at least 1 hour.
NOTE
The system must run for at least 1 hour before the temperature of the buffer or water is
measured. The length of time required for equilibration depends on the rotor and centrifuge starting
temperatures and the ambient room temperature.

After at least 1 hour, measure the temperature of the buffer or water using a thermometer or
thermocouple.
