Run the Auto Tune (Calibration) Procedure

Running the auto tune (calibration) procedure requires the use of software. Refer to the
NOTE
appropriate software guide (TRILUTION® LC Software or Masscape® Software) for additional
information and details.
1. Start the Auto Tune (calibration) procedure, which may take up to 30 minutes to complete.
2. Use a flow rate of 0.3 µL/min.
3. Select the tune file based on the ionization mode used: positive or negative.
● If using Masscape Software use scan settings of 50–1400 m/z, step 0.2 m/z, and scan
time of 1s.
4. confirm results from the Auto Tune (calibration) procedure.
● If the Auto Tune (calibration) procedure is successful, the settings will be automatically
saved for future sample analyses. Users may be prompted to keep or discard these
settings.
● If the Auto Tune (calibration) procedure fails, refer to the
Troubleshooting Table on page 72.

Flush the System

Gilson recommends flushing the system after the Auto Tune (calibration) procedure is
complete to remove any traces of the calibration solution. Follow the procedures below to
flush the SFI and spraychip to remove any trace of the calibration solution.
1. Disconnect the SFI plug and tubing from the SFI.
2. Manually flush the syringe and the infusion line with 50:50 (v/v) mixture of HPLc grade
methanol and water with no additives.
3. connect the PEEK tubing provided for the SFI to the input and waste line ports.
4. Attach the SFI to the Lc system and infuse 50:50 (v/v) mixture of HPLc grade methanol
and water with no additives with a make up pump flow rate of 1 mL/min for five minutes.
