QuantStudio ™ Real-Time PCR Software v1.6 does not require a normalization
calibration for the 96-well plate blocks or the 384-well plate block. For this version of
the software, normalization calibration is only required for the TaqMan
block.
After the software is upgraded, the instrument must be calibrated before starting a
run with a 96-well plate block or a 384-well plate block.

The feature to override a calibration can only be used for a data file with compatible
calibration data.
• A data file generated with a previous version of the software cannot be have the
calibration overriden after an upgrade to QuantStudio
v1.6.
• A data file generated with QuantStudio
have the calibration overriden if it is opened using a previous version of the
software.
The instrument requires regular planned maintenance by Thermo Fisher Scientific
service personnel to ensure that the instrument is operating within the installation
specification. The planned maintenance service can help you maximize your

The QuantStudio Real-Time PCR Software features an automated notification
system that alerts you of required calibrations. The feature will also prevent you from
operating an instrument that is out of calibration. For more information on setting the
