5.2 Fluorescence Microscope Mode

Fluorescence Microscope View consist of three different sections:
Profile Settings panel: Profile settings containing parameters used for scanning.
Microscope Control Panel: Live microscope movement functions
Main functions: Located at below the window header, these functions are identical in both
modes.

5.2.1 Profile Settings panel

Set Parameters to auto
Every parameter is set automatically during scanning.
Filter Settings
Compact table view of microscope filters used for Fluorescence scanning.
Legend of the table:
A: Actually used filter
Use: Filters used for viewing Live image and for scanning.
Auto Exp: When activated, the exposure time of the selected filter is calculated automatically
during scanning.
Exp. Time: If Auto Exp is inactive, exposure time of each filter can be defined manually.
Values such as Calc Exposure time in Microscope Control panel and those that are specified
manually are loaded in these fields. Value units can be the following: microsec, millisec, sec.
Exc %: The given value specifies the excitation intensity in percentage of the illumination unit.
DG (Digital Gain): This is camera-dependent, with Point Grey Grasshopper 3 the index value
ranges from 0 to 4, and with pco.edge 4.2 from 0 to 6. Give Index value by entering it in the
box or by selecting the value with the arrow buttons. If you raise the Index value, the
exposure time is halved by every step, and the image becomes noisier. To move between the
filters, click Previous Filter or Next Filter.
TW % : You can set the Exposure time weight (%) here (displayed under TW in the Filter
Selection frame).
F : Focus channel under the scanning
S : The selected channel will be the stitching channel under the scanning.
