MagicPrep NGS Instrument Functional Test
The protocol below outlines the steps necessary to run a functional library preparation test of the
MagicPrep NGS system.
When setting up the MagicPrep NGS system for the first time, it is useful to perform a functional
test in addition to the hardware test to confirm performance and set a baseline for your system.
A functional test may also be advised after a system error where a hardware failure was not
detected.
Kit: Revelo DNA-Seq Enz for MagicPrep NGS
Input: 100 ng human genomic DNA (NA12878, Coriell (https://www.coriell.org), Cat #: GM12878)
Number of PCR cycles: 6
6.6.1 Starting the run:
1. Follow the instructions provided in the Revelo DNA-Seq Enz for MagicPrep NGS user guide.
6.6.2 Quantify the libraries:
1. Remove the libraries from the MagicPrep NGS instrument.
2. Measure the library yield using a Qubit. Follow the instructions for measuring DNA
concentration on a Qubit.
6.6.3 Specifications:
The total yield should be ≥ 200 ng.
The coefficient of variation (%CV) across the 8 samples should be ≤ 20%.

Possible Cause
The temperature
measurement error with
the PCR/Thermal
module has occurred.
It is not possible to
continue the library
preparation.

If the error still exists, please contact Tecan
Customer Support.
Stop the library run
Follow the on-screen instructions to remove
the used reagent cartridge and sample deck
Switch off the instrument
Switch on the instrument to initiate the self-
test/diagnostic sequence.
Reload the MagicPrep NGS and try to
repeat the library preparation.
