If the reagent cartridge is not compatible with the flow cell, a message appears. Select Back to load a
compatible cartridge, or select Exit to return to the Home screen.

Close the reagent compartment door.

Select Next.
Start the Run
After loading the flow cell and reagents, review the run parameters and perform a pre-run check before
starting the run.
NOTE
The run starts automatically if the Start run after pre-run check option is selected in Run Settings. For more
information, see
Review Run Parameters

Review Experiment Name, Module Name, Read Type, Read Length, and Custom Primers. These
parameters are specified in the sample sheet.

When in Local Run Manager or Manual mode, select Edit to make changes to Read Type,
Read Length, or Custom Primers and then select Save.
NOTE
Changes to indexes must be done in the sample sheet. When in Sample Sheet mode, changes must be
made to the original sample sheet and then it must be reloaded.

Select Change Folder to review the folder locations.

Modify as needed and then select Save and Continue.

Select Next.
Change Folders
To change folder locations, select Change Folder and browse to a preferred location. Using this option from
the Review screen changes folder locations for all subsequent runs.
Review Pre-Run Check
The system performs a check of all run components, disk space, and network connections before starting
the run.
If any items do not pass the pre-run check, a message appears on the screen with instructions on how to
correct the error. For more information, see
When all items successfully pass the pre-run check, select Start Run.
