7.1 Emptying the Fluid Inlet Lines

Procedures to carry out after Cultivation
This chapter contains descriptions of procedures that have to be
carried out regarding the equipment after cultivation. This includes
the following:
Stopping the bioreactor, harvesting the culture (if necessary),
emptying the fluid inlet hoses ("Fluid Addition Lines") and
clamping off the silicone hoses of the reagent bottles
Disconnecting all cable and hose connections between the
basic unit of Labfors 5 Cell and the single-use vessel:
Disconnecting the silicone hoses from the fluid inlets
("Fluid Addition Lines") on the single-use vessel
Disconnecting sensor cables from the sensors
Removing the temperature sensor (Pt-100) from the
thermowell in the single-use vessel
Disconnecting the gassing hose on the equipment from
the sparger line on the single-use vessel
Unplugging the heater pad
Removing the motor
Removing the heater pad
Removing the sensors from the single-use vessel
Sterilising the sensors and reagent bottles with pump heads in
the autoclave
Cleaning the sensors, reagent bottles, pump heads and
heater pad
Descriptions of maintenance and disposal of the single-use vessel
can be found in the documentation from the manufacturer Merck
Millipore™ and must be adhered to.
The operating manual of the bioreactor Labfors 5 must be read and
its content understood before beginning any work.
When the cultivation is finished and the bioreactor is stopped:
If needed, sterile harvest of the contents of the single-use vessel
can be carried out first. Then the fluid inlet lines ("Fluid Addition
Lines") must be emptied.
A possible sterile harvest has to be carried out according to
internal regulations and instructions. This procedure is not
described in this document.
