6.1.3 Running Gradient Protocols
The temperature gradient will appear in the graphical representation of the
protocol. The maximum temperature, minimum temperature, and thermometer
icon will be displayed for the gradient segment. This graphical presentation of the
protocol appears in the run screen while the protocol is running.

6.1.4 Converting a Gradient Protocol
The MyCycler Thermal Cycler allows the user to convert an existing gradient
protocol to a non gradient protocol. To use the conversion feature, select the
F1-Option to go to the Protocol Library. Select the gradient protocol that will be
converted. From the Choose Operation Menu select Convert Gradient. A menu
will appear that contains a temperature for each row of the gradient. Select the
conversion temperature from the Choose Row Menu. Save the protocol with a
unique name.
Please note that when a gradient protocol is converted to a non gradient
protocol the ramp rates associated with the gradient protocol are transferred to the
non gradient protocol. This is done so that the conversion of a protocol will produce
an identical result. A black triangle in the upper left corner of a protocol step
indicates that the ramp rate has been changed. When a step containing an altered
ramp rate is highlighted the actual rate is shown below the protocol. For more
details on changing ramp rates please review Section 5.4.3 in this manual.
