Flushing the Sample Transfer Line, Sample Tube, and API Probe

After operating the TSQ system, verify the instrument is ready for a period of inactivity by
doing the following:
• Flushing the Sample Transfer Line, Sample Tube, and API Probe
• Placing the System in Standby Condition
• Cleaning the Ion Sweep Cone and Ion Transfer Tube
• Emptying the Solvent Waste Bottle
Thermo Fisher Scientific recommends flushing the sample transfer line, sample tube, and API
probe at the end of each working day (or more often if you suspect they are contaminated) by
flowing a 50:50 methanol/distilled water solution from the LC through the API source.
 To flush the sample transfer line, sample tube, and API probe
1. Wait until data acquisition, if any, is complete.
2. To display the EZ Tune window, from the Windows taskbar choose Start > All Programs
> Thermo Instruments > TSQ > TSQ Tune.
3. From the EZ Tune window, choose Control > On (or click the On/Standby button) to
turn on the voltages and gas flows to the API source.
• If you are operating in APCI mode, go to step 4.
• If you are operating in ESI mode, go to step 5.
4. Set up the APCI source:
a. In the EZ Tune window, click on the Ion Source button to display the Ion Source
Devices dialog box.
b. Set the APCI vaporizer temperature to 500 °C:
Select Vaporizer Temperature in the device list, and then type 500 in the box and
click Apply.
c. Set the sheath gas flow rate to 30:
Select Sheath Gas Pressure in the device list, and then type 30 in the box and click
Apply.
d. Set the auxiliary gas flow rate to 5:
Select Aux Valve Flow in the device list, and then type 5 in the box and click Apply.
e. Set the spray current to 0:
Select Spray Current in the device list, and then type 0 in the box and click Apply.
Go to step 6.
