If there are no standard replicates. SmartSpec 3000 will direct you to enter
the concentration of each standard and then to measure the absorbance of each
standard.
Enter concentration of
standard #1:

Use the number keys to input the concentration of standard #1 and press Enter.
You may enter concentrations with as many as eight significant digits, but all results
are reported with four significant digits because SmartSpec 3000 absorbance data are
measured to four significant digits.
Insert Standard #1 and
press Read Sample

Place the cuvette with standard #1 in SmartSpec 3000 and press Read Sample.
SmartSpec 3000 will continue to prompt for all subsequent standards until the last
one is read. Before each standard is measured, you will be asked to specify the
concentration of the standard. The units of concentration for all standards must be the
same as those specified before the first standard was read.

press Read Sample

Insert Standard #9 and

When collecting data for construction of the standard curve, SmartSpec 3000 does
not display the absorbance data, but you may view it at any time by pressing Abs.
Standard #1

Press the right arrow key to return to the assay.
Once all the standards are read, SmartSpec 3000 will calculate a standard curve. Skip
ahead to C.
ii. If there are standard replicates. If you indicate that one or more of the
standards are read in replicate, then you will be asked if there is the same
number of replicates for each standard.
