Acquiring fluorescence images

3. To acquire an image without background, adjust the intensity parameters. The intensity should stay low (<20-30) (again to minimize
photobleaching). Now, reduce the gain until the background disappears. The maximum gain to be used can be up to 40%.
Increase the exposure time if you need to see more details of the image but try to keep it as low as possible (100 ms or lower = very
good; 200 ms = good) (Figure 32).
