    Thank you for choosing the QIAcube HT. We are confident it will become an integral part of your laboratory. Before using the QIAcube HT, it is essential that you read this user manual carefully and pay particular attention to the safety information.

  • Page 12: Requirements For Qiacube Ht Users

    Requirements for using dedicated QIAcube HT kits on the QIAxtractor To process dedicated QIAcube HT kits on the QIAxtractor instrument, QIAcube HT Software version 4.17 or higher is needed together with the Accessories Pack, QXT (see Appendix C – Ordering Information).

    Safety Information Before using the QIAcube HT, it is essential that you read this user manual carefully and pay particular attention to the safety information. The instructions and safety information in the user manual must be followed to ensure safe operation of the instrument and to maintain the instrument in a safe condition.
  • Page 14: Electrical Safety

    The robotic arm moves during position calibration while the instrument lid is raised. Never click any buttons while parts of your body are within the instrument workspace. In case of emergency, switch off the QIAcube HT at the power switch at the rear of the instrument and unplug the power cord from the power outlet.

    If liquid has spilled inside the instrument, switch off the instrument, disconnect it from the power outlet, and contact  QIAGEN Technical Services. If the instrument becomes electrically unsafe, prevent other personnel from operating it, and contact QIAGEN Technical  Services; the instrument may be electrically unsafe when: The line power cord appears to be damaged.

    WARNING Moving parts Avoid contact with moving parts during operation of the QIAcube HT. Under no circumstances should you place your hands under the robotic arm when it is lowering. Do not attempt to move any plates or tubes while the robotic arm is operating.

    Do not place any items on the robotic arm.  Select Pause, located in the toolbar of the QIAcube HT Software, to pause the instrument before removing plates or tubes  from the worktable. Wait until the robotic arm has completed the current movement — this may take up to 10 seconds.

    General Description The QIAcube HT delivers high performance and reliability, enabling purification of high-quality nucleic acids from 8–96 samples per run, in increments of 8 samples. Innovative instrument features ensure reliable purification and minimize the risk of contamination. The QIAcube HT is a fully integrated solution and provides reagents, plasticware, and optimized protocols for nucleic acid purification from virtually any type of sample.

    The UV lamp does not irradiate certain areas, including the pipetting head, due to construction constraints. The UV lamp is enabled and time of exposure is set within the QIAcube HT Software. For more details, see Section 5.18.3, page 133.

    Tip chute Tip chute. Tips used on the worktable are discarded through the tip chute. Located at the rear-left of the QIAcube HT, the tip chute is held in place by a locating block, allowing easy placement and removal for cleaning.

    Riser block EMTR Vacuum chamber, channeling adapter, and riser block EMTR (unassembled). The QIAcube HT vacuum chamber is composed of two separate compartments for waste (left) and elution (right). This configuration is one of many measures that minimize sample cross-contamination during purification.

    The QIAcube HT is operated via a computer. A laptop with the correct specification for operation of the QIAcube HT and QIAcube HT Software is supplied as part of the QIAcube HT System. However, if a different computer is used to operate the QIAcube HT or to run the QIAcube HT Software, then the following desktop or laptop hosting systems are recommended.

    Software installation Note: QIAcube HT Software must be installed by a user with Windows administrator rights (see Section 4.5.1, page 37) before the instrument can be connected to the laptop for the first time. Installing the QIAcube HT Software Note: The software will usually be installed on the laptop by a QIAGEN Field Service Specialist on the day of instrument installation, if the QIAcube HT was purchased with an installation pack.

    The QIAcube HT uses USB for connection to the computer. The laptop which is supplied with the QIAcube HT should be installed via USB. Note: Connect the computer to the QIAcube HT either via the USB or via the RS-232 interface. Do not make both connections at the same time.

    Configuring the COM port The COM port used to connect the computer to the QIAcube HT must be correctly configured in the software to enable the software to communicate with the QIAcube HT instrument. By default, the software is configured to use COM1, and this corresponds to the first RS-232 port on the system.

    QIAcube HT Software The QIAcube HT Software provides a user-friendly, easy-to-use interface for configuring and running protocols on the QIAcube HT instrument. When started, the software automatically detects if the instrument is switched on and connected properly. The software displays a layout that corresponds to the setup of the instrument worktable.

    Protocols can be specified and saved in the “Virtual” mode to create templates for subsequent runs. This feature enables the user to create protocols on any laptop that has QIAcube HT Software installed. The template can be transferred to the laptop connected to the instrument, minimizing the time required for working on the laptop connected to the instrument.

    Identifying the instrument serial number Each QIAcube HT is identified by a unique serial number, which is displayed on the serial number badge at the rear of the instrument. The serial number is also stored electronically in the instrument, allowing the instrument to be identified by the software.

    Sample names can be easily copied from a spreadsheet by copying the column that contains the sample name information. The copied entries are pasted to the QIAcube HT Software by right-clicking a cell and selecting the “Paste to current cell” item from the menu that appears.

    User management User management allows the definition of different access rights to various data and functions within the QIAcube HT Software. User management functions are accessed from the menu bar (Options > User Management). User identification Unlike software that requires separate login authentication, the QIAcube HT Software user is defined as the current Windows user logged on the laptop on which the software is installed.
  • Page 86 Upon restarting the software, only Administrators and users assigned to the “Manager”, “Designer” and “Operator” groups can operate the software. When attempting to start the QIAcube HT Software, users that are not assigned to a user groups are denied access to the software.

    Enabling access control Upon installation, the QIAcube HT Software user access control features are disabled and all users have “Manager” access (see the previous section for user group descriptions). A Manager enables the user access control with Options > User management > Enable access control.

    Alternatively, select Options > User management > Change Current User. The “Change User” window appears. The new user must enter their username and password, and click OK to log on.     The use of a joy pad (a game controller that has only buttons and no stick) can make position calibration significantly easier. A USB joy pad can be connected to the laptop that controls the QIAcube HT. During calibration, the user controls the robotic arm movement with the joy pad.
