3. Connect the OTKB-FL segment to the beam expansion segment as shown in the Figure 3.
4. Attach the camera segment onto OTKB-FL as shown in Figure 3.
5. Adjust the TR50 post to keep the collimator segment horizontal.
6. Insert the liquid light guide into the LLG3A1-A connector and lock this connection.
7. Power up the light source
8. Wait for the lamp to warm up and open the shutter.
9. An excitation light source should be seen emitting from the objective lens. Adjust the two KCB1 mirror
mounts in the OTKB-FL module to maximize the light emitted from the objective lens.
10. The complete setup should be similar to Figure 3.

Additional Alignment Tips

If the system is slightly off aligned, please follow the instructions below to align your OTKB-FL.
1. All adjustments for the fluorescence module must come from the two KCB1s in the fluorescence module.
Adjust the two KCB1 to make sure the light output from the objective lens is maximized.
2. Please check the collimator segment is horizontal. Adjust TR1 height to level the collimator segment.
3. Adjustment of the KCB1s in vertical segment will affect your trapping laser beam path.
4. Adjustment of the KCB1 in the camera segment will not affect light going into the objective lens but will
affect the region of interest for imaging.
5. Use fluorescence micro-beads as the sample. You can test your detection system by turning off the white
light source (at the top of the vertical segment) and observe sample area on camera. If the alignment is
correct, you should be able to see the fluorescent beads on the camera.
