Zooming In and Out. You can zoom in and out of the hemocytometer image on the
touchscreen. To zoom in, spread your fingers on the image as shown in the example
that follows. To zoom out, pinch the image.
Changing Contrast Parameters. For the channel on display, you can change the
contrast parameters as needed:

Low Contrast
For the selected channel, set the low contrast limit for the images

on display.
High Contrast
For the selected channel, set the high contrast limit for the images

Drag the sliders to adjust the low (bottom slider) and high (top slider) contrast
limits to a desired location. You can also use the plus (+) and minus (–) signs to
refine your contrast limit.

Adjust each channel as needed.
Changing the Exposure Time. For each channel, you can re-adjust the exposure
time as needed, and then tap Image. An increase exposure time increases the signal
strength, and a decrease in exposure time decreases the signal strength.
IMPORTANT
• Be certain that you always tap Image after making any parameter adjustments so
that the hemocytometer data is up-to-date.
• If you see a message stating that the focus cannot continue, increase the exposure
time and tap Refocus.
(Optional) View Hemocytometer Data Outside the Run
If you had inserted at least a 3 GB USB key into the USB portal on Polaris before
performing the Hemocytometer step, you can view the hemocytometer data by
opening the MS Excel HemocytometerData.xlsx file on the USB drive.
To view hemocytometer data outside the run:

At the bottom-left of the Select an experiment to run screen, tap the Fluidigm logo:
