Introduction Milliflex Quantum System is a rapid fluorescence based technology for fast quantitative detection of contaminants over a broad range of filterable matrices. This easy to use and simple system can detect viable microorganisms filtered on a membrane down to one CFU per sample. The non-destructive method allows the identification, by any method, of any microorganisms detected during the initial fluorescence reading.

Installing the Milliflex Quantum Reader The Milliflex Quantum Reader weighs 4.4 kg (9.7 lb) and is designed for use on a laboratory workbench. Remove all components and their accessories from the boxes. Remove the reader from the protective bag and place it on a flat, stable, horizontal surface.

Connect the end of the power cord to the power outlet on the reader . LED Power Indicator Connect the power cord to the external power supply. Connect the external power supply to a power source. The system will start up. The auto test sequence will run and verify that the Milliflex Quantum Reader is ready to use.
Installing the Membrane Transfer Tool NOTE: The Membrane Transfer Tool should be used in a microbiological safety cabinet or laminar flow hood. The Membrane Transfer Tool and seal may be autoclaved for 30 minutes at 121°C or for 10 minutes at 134°C. Do not autoclave the membrane transfer tool with the seal installed.

Press on the membrane transfer tool to lock it in place If the Milliflex PLUS Pump is OFF, turn it on. Run the tare cycle on the pump as described in the Milliflex PLUS Pump User Guide. The membrane transfer tool is ready to use.
Preparing the Removal Rack NOTES The Removal Rack should be used in a microbiological safety cabinet or laminar flow hood. The Removal Rack may be autoclaved for 30 minutes at 121°C or for 10 minutes at 134°C. The Removal Rack is ready to use and requires no preparation. Installing the Camera NOTE Do not place the reader in the stand if the camera will be installed.

All screenshots shown are examples and may vary from one computer to another. Switch on the computer. Log on with an administrator account. Insert the installation disc into the CD drive of the PC. Using the file explorer browse to the CD-Rom and double-click the “Setup. exe”...

A new window is displayed, listing the components required to complete the Milliflex Quantum software installation. Click Install. When the operation is completed, click Next. A message indicating that the software is now ready to be installed is displayed. Click Install.

A message indicating that the computer must be restarted in order to complete the installation of the software is displayed. Save and close all the other applications. Then, select Yes, restart the computer now and click Finish. After the computer has restarted the QuantumSpotCounter icon is displayed on the desktop.

Preparing the Reagent Kit Precautions • Do not use any expired kit component. • Remove the kit from the refrigerator one hour before starting the reconstitution. • Reconstitute under a laminar flow hood or a microbiological safety cabinet. • During the seven days shelf life, do not let the staining solution remain at room temperature for more than four hours per day.
Using the Milliflex Labels (optional) Record the test information on the Milliflex Labels. NOTE: The Label Printing software, available on the Millipore website, allows to print data on the Milliflex labels. Please refer to the Label Printing Software User Guide. Before the sample filtration, outside the laminar flow hood or the microbiological safety cabinet, apply the label to the top of the solid media cassette.
Filtering Perform the sample filtration as described in the Milliflex PLUS Pump User Guide. Perform the membrane transfer on a Milliflex solid media cassette,and the breaking-off of the funnel either manually or with the MilliSnap™ System. Incubating Place the Milliflex membrane on the Milliflex solid media cassette in the incubator, with the funnel cover facing down.

With a pipette, add 2 mL of staining solution on the center of the pad of the Milliflex liquid cassette NOTE Shake the staining solution before the first staining of the day. Do not touch the pad when adding the staining solution. Using the Removal Rack, separate the Milliflex membrane from the Milliflex solid media cassette.

Push on the lever of the membrane transfer tool to remove the Milliflex membrane on the Milliflex liquid cassette from the tool. Incubate the Milliflex membrane on the Milliflex liquid cassette for 30 minutes at 32.5 °C + 2.5 °C with the funnel cover facing down. NOTE The Milliflex membranes on the Milliflex liquid cassettes should not be stacked in the incubator to ensure that all membranes are incubated...
Installing the Milliflex Membrane in the Reader After the staining step, remove the Milliflex membrane on the Milliflex liquid cassette from the incubator. Verify that the reader is on and ready to use. Open the reader drawer and place the Milliflex membrane on the Milliflex liquid cassette in the drawer.
Reading the Membrane Reading the Membrane Manually NOTE : Aligning the Milliflex membrane grid with the drawer will make counting easier. Once the reader drawer is closed, the reader lights the Milliflex membrane. The screen displays the counter. Push the counting button to increase the count. NOTE : After 10 minutes without opening the drawer, the LEDs inside the reader switch off and the system enters standby mode.
Reading the membrane with the Milliflex Quantum Camera Starting the Milliflex Quantum Spot Counter Software Double-click the icon on the desktop. The software opens. NOTE If the Milliflex Quantum Camera is not connected, the message below is displayed before the opening of the software. NOTE The camera must be connected to the same USB port during installation and use.

The Milliflex membrane image is displayed. Counting Place the mouse on the Milliflex membrane image. A cross appears on the screen. Place the cross on a spot by moving the mouse. Using the left button of the mouse, click the spots to mark them. The counter increases by one with each click.

NOTE When the FullScreen mode is activated, the counter value and the zoom factor are displayed in the toolbar menu, at the top of the screen. Click the FullScreen menu to switch to the normal mode. Pen size Menu The pen size can be changed by clicking the Pen size menu. Four types of pen are available.

Count the spots in the outlined segment. The extrapolated number of spots for the entire Milliflex membrane is indicated at the top of the screen. Click Extrapolation to quit this function. Reset Menu Click the Reset menu to erase all the spots marking and reset the counter.

To print the image with counting marks, select Print with overlay. Opening a Saved Image Click the File menu. Select the Open. Once the image opens, the image functions are available. NOTE The Milliflex Quantum Camera does not need to be connected to open and work on a saved image.
Reincubation After the reading of the Milliflex membrane separate the Milliflex membrane from the Milliflex Liquid Cassette using the Removal Rack. (Refer to the Removal Rack User Guide). Keep the cover on the membrane. Remove the cover of a new Milliflex solid media cassette. Place the Milliflex membrane onto the new Milliflex solid media cassette.
Maintenance Cleaning recommendations Disconnect the reader from the power supply before cleaning or servicing. Do not spray any cleaning products, solvents or abrasives on and in the Milliflex Quantum Reader and Camera, as these could damage the instrument. Membrane Transfer Tool Before the first transfer of the day, gently clean the surface of the membrane transfer tool with a non woven wipe soaked in one of the products listed in the Cleaning recommendations section. Before each transfer, wipe the surface with a dry wipe. After the last transfer of the day, clean the seal with a non woven wipe soaked in one of the products listed in the Cleaning recommendations section.
Let the tool dry on an absorbent cloth. Autoclave the tool and the seal at 121°C during 30 minutes or at 134°C during 10 minutes. NOTE Remove the seal from the tool before autoclaving. Reinstall the seal on the membrane transfer tool. Removal Rack Disassemble the Removal Rack as described in the Removal Rack User Guide and clean using a non woven wipe soaked in one of the products listed in the Cleaning recommendations section.
Semi-Annual Servicing Replace the seal on the membrane transfer tool Annual Servicing A complete check of the Milliflex Quantum System should be performed by an authorized Millipore Technician.
