Section 4: Experimental Tutorials

Once the VP-ITC is setup and running properly, new users may proceed directly to the tutorials in
this section for a 'hands-on' approach to learning the instrument. When more detail is required
about the software or experiment the user may refer back to sections 2 or 3.
The water-water tutorial provides the user experience in loading the pipette and cells. Obtaining
small and repeatable water into water injections will serve to establish faith in the ITC and
techniques of operation. The methanol-water tutorial will closely mimic a typical titration
experiment with respect to the size of the heats. Finally the Rnase-2'CMP provides a real
experiment to be analyzed and compared with the results obtained with your instrument at
MicroCal.
The following experimental tutorial is designed to acquaint you with the basic features of both the
hardware and software of the VP-ITC instrument, as well as to provide experience with several
manipulations that must be mastered in order to get the highest quality data from your instrument.
Rather than beginning experimentation on the VP-ITC using precious biological samples, we
strongly suggest that each new user of the VP-ITC instrument complete the following tutorials
first
