Insufficient negative Open vacuum regulator

Manifold top not seated

Unused CAP luer fittings
not closed completely

chips and cracks)

Possible Solution
Close vacuum regulator
by turning completely
clockwise
Ensure uniform contact
between manifold top
and base
Use A stage for
waste collection
Press down gently on
plate or CAP to seal
Close unused luer
fittings completely with
distilled water
Press gasket back into
groove
Replace vacuum
manifold or CAP
If closure of wells or
columns (e.g. with
sealing tape restores
negative pressure, no
corrective action
needed.
Press down gently on
plate or CAP to seat
distilled water
