Move the objective vertical movement lever to the
left so that the objective tip touches the surface of
the water over the specimen.

To perform the parfocal correction, use the
parfocal correction knob on the sliding nosepiece.
With this focusing operation, a confocal position
for the two objectives can be attained.

Adjust the brightness with the brightness control
knob on the power supply or on the optical fiber
light source.

Stop down the field diaphragm with the field
diaphragm control so that it circumscribes the
viewfield.
Refer to Section 6 in Chapter 3, "Field
Diaphragm."

Stop down the condenser aperture diaphragm
with the condenser aperture diaphragm lever to
about 70 to 80% of the numerical aperture of the
objective.

Start the microscopy.
