Priming procedure
ÄKTA ready XL Operating Instructions 29281616 AE
TeDIS Valid on Date Printed
This instruction sets column and filter to bypass, and ramps up the pump adequately
for priming.
Note:
The air trap is set to inline as default and can be used to visually detect air
bubbles. If required, set the air trap to bypass prior to running pump pri-
ming.
The instructions below are a example procedure for UNICORN software.
For other control softwares, refer to the software documentation to create a similar
procedure.
Step
Action
1
Make sure the relevant inlet or inlets each are connected to a tank contain-
ing a suitable solution.
2
Open the connected inlet valve located furthest from the pump using the
instruction Valves.
Tip:
Inlet A6 is the inlet located furthest from the pump, Inlet A5 is the second
furthest etc.
3
Fill the flow kit tubing from the connected inlet located furthest from the
pump using Pump →ManFlow set to approximately 30% of the maximum
flow rate.
4
Fill the flow kit tubing of each of the connected inlets, by opening the sec-
ond furthest inlet valve, then the third etc. until all connected inlets are fil-
led. Make sure that the priming liquid reaches the pump. This procedure
may take > 20 sec.
5
Open inlet Inlet A1.
Tip:
Inlet A1 is used for prime flow as it leads straight to the pump.
6
Select PumpPrime and set the parameter to Run.
Result:
The pump gradually ramps up to maximum priming speed to remove air
from the flow path.
7
Run the pump until no air is detected, i.e the tubing leading to the air trap
no longer vibrates, and the air trap content is free from bubbles. This pro-
cedure may take more than 1 minute.
