    Basic Information Basic Information Notes on the manual The user manual describes the following two models of the qTOWER 84 series: qTOWER  qTOWER 84 G  In the text below these devices are collectively called qTOWER 84. Differences be- tween the models are explained in the corresponding section.
  • Page 6 Basic Information Analytik Jena AG offers a number of color or FRET modules of which up to six can be installed simultaneously in the device. Replacement and retrofit of color or FRET mod- ules can be easily accomplished. qTOWER³ 84 is an open platform for real-time PCR and supports both intercalating dyes as well as individual samples and kits of various manufacturers.

    Function and setup The fluorescence dyes then emit a light of longer wavelength that is bundled through the lenses in the shuttle system and passed via the optic fibers back to the color mod- ules. In the color modules the light passes the beam divider followed by two emission filters and is then transferred to the photomultiplier (CPM) for detection.

    Function and setup PCR thermocycler The qTOWER³ 84 is equipped with a thermal block with 384 wells. The thermal block consists of very good heat-conducting aluminum. This achieves a high temperature homogeneity and uniformity in combination with heating rates of up to 4 °C/s and cool- ing rates of up to 2 °C/s.

    Commissioning The qTOWER³ 84 is opened by folding back the upper part and the cover for the sam- ple block it contains and the fluorescence photometer. To this end press the handle in until the lock disengages with a click and the upper part gently snaps open. The upper part can then be folded back from the handle.

  • Page 30 Commissioning Place the device on its side. Check the set operating voltage. Use the voltage selection switch (Fig. 7 S. 26) to adjust the operating voltage to the mains voltage. Adjust the switch using a screwdriver or a coin so that the arrow points in the di- rection of the mains voltage available on site.

    PCR plates with 384 wells. These and other consumables can be ordered from Analytik Jena AG. After pipetting the PCR samples into the wells these must be sealed with a transparent optical adhesive film (sealing film). The optical transparency of the films affects the fluo- rescence signal directly.

Any unauthorized interference limits warranty entitlements. If the device exhibits any faults or details, please contact the Analytik Jena AG customer service immediately.

    Maintenance and care Installing color modules Depending on the delivery state of the qTOWER³ 84 or when purchasing additional color modules, it might be necessary to install the color modules in the device. The top device hood must be removed for this purpose. NOTICE Sensitive optics! Do not remove the interior hood, because there are underneath sensitive optical elements.
  • Page 38 Maintenance and care  The pivotable holder for the color modules is now accessi- ble. Note the code on the color module and the position in which the color module is to be installed. Place the color module onto the free position with the pin pointing down.
  • Page 39 Maintenance and care 11. Rotate the wheel with the installed color modules and check whether the color modules grind against the lid. If this is the case, loosen the screw connection of the color module, check its position and retighten the screws. NOTICE Do not attempt to force a jammed color module into the correct position simply by tighten- ing it further.
