In order to ensure electromagnetic compliance, connect peripheral devices only with double-shielded cables and only use the power cable and the keypad included in delivery.     CAUTION! Two persons are required to lift the instrument. Take care to avoid personal strain or injury. The instrument is heavier at the front and should be stabilized while lifting it. Miltenyi Biotec accepts no liability for potential injuries sustained during lifting and/or movement of the device.

    Assembly 2.6.1 Install the fluid bottles Operating the instrument requires Running Buffer, Washing Solution, and Storage Solution. One bottle is required for waste collection. Always operate the instrument with ready-to-use MACS® Buffers and solutions only. The bottles, closures, tubing and fluid sensor cables are color-coded for easier handling. 1 Install only one fluid bottle at a time.

    7 Remove the protective cap and connect a hydrophobic air filter to the appropriate connector of the fluidics system on the front of the instrument. 2.6.2 Install the Single tube rack The instrument is delivered with the Single tube rack. The Single tube rack is compatible with all standard flow cytometry tubes (1.5 mL, 2 mL, and 5 mL).

    3 Set the following parameter: Standby timer, the amount of idle time before shutdown begins. Shutdown timer, the incubation time during the shutdown procedure (5 minutes is recommended) Needle priming time, to avoid that the needle runs dry (20 seconds are recommended) 4 Set the Shutdown default behavior to either Analysis mode or Instrument off .

    MACSQuantify™ Software. It is highly recommended that hardware calibration is only performed by an administrator who has been trained by Miltenyi Biotec. Refer to the MACSQuantify™ Software manual and/or the help function of the MACSQuant Instrument for an introduction to the MACSQuant Instrument user interface and a detailed description of the instrument settings.
  • Page 64 3 Manually adjust the robotic needle arm in order to position the uptake needle over the center of the port. 4 Gently lower the robotic needle arm manually into the port until the uptake needle touches the bottom of the port. Do not bend the needle or insert it diagonally. 5 Click Save calibration.

    4 Place the MACSQuant X 5 Rack onto the Orbital Shaker and place an empty 5 mL tube in position D6. 5 Click Start calibration . The robotic needle arm will automatically move forward and insert the uptake needle into the tube in the position D6 of the rack.

    4.1.4 Calibration of the 96-well plate 1 Click on the Tools tab on the sidepane of the Software and check the Calibrate uptake unit box. Red closed circles indicate that the uptake components are not calibrated. 2 Click the Experiment tab in the sidepane of the Software and select the 96-well format from the Rack pull- down menu.

    2 Click the Experiment tab on the sidebar of the MACSQuantify Software and select Chill 96 rack from the Rack pull-down menu. If the MACSQuant X 5 Rack is not selected, the MACSQuant Instrument will display a prompt screen.

    This process is automated as an express mode once a month. 1 Install either the single tube rack or the MACSQuant X 5 Rack. If using the MACSQuant X 5 Rack, select a single position.
  • Page 69 3 Click the Start button. 4 Ensure that buffer bottle is full and the waste bottle is empty. Follow the instruction and click Continue . 5 After the volume calibration has completed, the status of the calibration is indicated in the upper right corner.

    Live Support MACSQuant® Live Support is a real-time diagnostic service provided by Miltenyi Biotec Technical Support. Highly trained MACSQuant Specialists can be reached in real-time to assist with any queries you may have. A webcam is provided for communication with the MACSQuant Live Support, which is attached via USB connection to the MACSQuant Instrument.

    To avoid clogging due to deposition of dust, exchange filters once a year. Please use only the hydrophobic air filters from Miltenyi Biotec. 1 To exchange the air filter of the fluidics system, remove the Single tube Rack if installed.

    Before exchanging the sample needle, make sure that the needle is in its home position (that is the needle is hidden within the needle arm). Figure 7.1: Needle arm and needle holder of the MACSQuant X. 1 Switch off the instrument.
  • Page 79 4 Unscrew the knurled screw of the sample tubing. Make sure not to loosen the inlay. 5 Remove the sample tubing and replace it with new tubing. 6 Insert the inlay into the needle holder. 7 Tighten the knurled screw.

  • Page 80: Exchange The Sample Tubing

    8 Insert the needle back into the slide in the z-axis. Do not bend the needle. 9 Snap the needle holder back into the needle arm. 10 As the needle has been removed from the needle arm, its position might have been changed. Check the position and recalibrate if necessary.
