TIRF microscopy
In TIRF microscopy, images are captured by TV camera and observed on the monitor.

Close the epi-fl attachment or the epi-fl cassette holder shutter.

Rotate the excitation method switching ring of the epi-fl cassette holder to select a hollow
without the filter block.

Rotate the excitation method switching ring of the TIRF cassette holder to switch to a desired
excitation method.

Close the eyepiece shutter and switch the microscope's optical path to a port to which the TV
camera is attached.

Use the shutter switch of the shutter control unit to open the motorized shutter of the desired
laser.

Use the field diaphragm open/close knob to fully open the field diaphragm.

If laser-focal position has not been fully adjusted, perform laser- focal position adjustment for
the objective in use by turning the focal point adjustment knob until the laser beam emitted
from the objective is collimated.

Never attempt to look into the laser beam. (FDA CDRH: Class II, EN60825-1: Class 2,
IEC60825-1: Class 2)
The objective emits a powerful laser beam. When adjusting the point adjustment knob or
condenser lens locking screw, never attempt to look into the objective or the areas near the
objective from above or below the stage as the lens emits a powerful laser beam toward the top
and bottom sides of the stage. To protect your eyes and skin from exposure to laser beam
reflections, never place or mount any reflective materials on the stage, in the room, or on the
chamber ceiling.

Adjust the laser position adjustment knobs to provide TIRF illumination.

Never attempt to look into the laser beam. (FDA CDRH: Class II, EN60825-1: Class 2,

The objective emits a powerful laser beam. When adjusting the laser position adjustment knob,
never attempt to look into the objective or the areas near the objective from above or below the
stage as the lens emits a powerful laser beam toward the top and bottom sides of the stage. To
protect your eyes and skin from exposure to laser beam reflections, never place or mount any
reflective materials on the stage, in the room, or on the chamber ceiling.
9. Adjust the field diaphragm open/close knob and the field diaphragm centering knob so that
only the required area is illuminated.

Turning off power

Follow the steps given below to turn off power to your equipment.

Switch off the key switch on the air-cooled argon laser remote controller.
Leave the laser unit standing for approximately 15 minutes, then check that it is sufficiently
cool. Switch off the key switch and the power switch on the power unit.
