INTRODUCTION General Description The Labconco Auto Densi-Flow is a single apparatus for both depositing a preformed gradient smoothly and continuously and gently removing centrifuged sample layers. The instrument obtains a high degree of resolution and can deposit or remove samples within 0.1 mm (0.004 inch) of the gradient surface.
A nylon strip is placed over the stainless steel rollers and is clipped in position at the block on the front edge of the top of the Auto Densi-Flow. This nylon strip should not be tight. It should bow out on the output side of the pumping head.
INSTALLATION Installation Pump 1. Remove the pressure bar. 2. Loop the tubing over the rollers with the nylon strip between the roller and the tubing. 3. Replace the pressure bar and tighten it slightly with the swivel clamp. 4. Loop the tubing around the 2 pins located in the semi-circular cutouts in the block retainer located in front of the pumping head.
NORMAL OPERATION Pump 1. Immerse the inlet end of the tubing (if depositing) or the end of the probe (for removal) into a suitable liquid. 2. Set the speed control to #9 and turn the switch to forward or reverse, as required. 3.
NORMAL OPERATION probe is placed just below the desired band. A syringe connected to the probe through a short length of tubing will usually be more useful for withdrawing the band than the pump. If more than one band has to be withdrawn, start with the uppermost band. After it is taken out, depress the override button to the next band, and so forth until all the bands of interest have been drawn out.
MAINTENANCE Care and Maintenance After completion of a run, rinse the probe with distilled water with the pump to prevent sucrose or any other material from crystallizing or fouling the probe and connecting tubing. Special care is required after use with cesium and other salt gradients because they can permanently damage the probe.
