Flow-Count (SPT) Method

The Flow-Count (SPT) Method uses Flow-Count Fluorospheres and a flow cytometer to determine
the absolute counts with the following formula:
Absolute Count (cells/μL) = (Count of Positively Stained Cells of Interest ÷
Count of Flow-Count Fluorospheres) x Flow-Count Fluorospheres Assayed Concentration
This Single Platform Technology (SPT) method is automated using the Navios software and requires
all of the following:
• Flow-Count Fluorospheres must be used.
• The Assayed concentration of Flow-Count Fluorospheres must be entered in the Absolute Count
Calibration dialog box and selected in Acquisition Manager when running a protocol that
includes a CAL region. See
• The number of fluorospheres counted must be 1000.
Note: A total white blood cell count is not necessary for determining an absolute count for the cells
of interest if using Flow-Count Fluorospheres.

Dual Platform Method

The Dual Platform Method calculates absolute counts by combining results from hematology and
flow cytometry and using the following formula:
Absolute Count (cells/μL) = Total White Blood Cell Count (cells/μL) x %Lymphocytes x
%Positively Stained Cells of Interest ÷ 104
Absolute counts are obtained using the Report function of the Navios Software. For information on

Setup Mode

During Setup mode the workspace is a rolling display that is updated up to three times per second.
The maximum number of events can be set on the
incoming data is not saved.

Acquisition

During data acquisition, the plots are updated in real time. When one plot is displayed with statistics
underneath, the statistics are also updated in real time. Unlimited one or two parameter plots are
available for any given sample.
One-parameter plots have 1,024-channel resolution.
Two-parameter plots have up to 512- x 512-channel resolution.
