Intended Use
Thermo Scientific™ Oxoid™ Schaedler Anaerobe Agar with Sheep
Blood (with Haemin and Vitamin K1) is a non-selective medium for the
isolation of obligate and facultative anaerobic bacteria.

Summary and Explanation
Schaedler, Dubos and Costello
isolation of aerobic and anaerobic microorganisms from the gastro-
intestinal tract of mice. Mata, Carrillo and Villatoro
formula in their studies on anaerobic human faecal microflora. This
modified formula has been used in Oxoid Schaedler Anaerobe Agar
with Sheep Blood (with Haemin and Vitamin K1). The medium is used
as a non-selective, highly nutritious agar for the isolation of
anaerobes.
Principle
Tryptone soya broth and special peptone supply nitrogen and other
nutrients necessary for growth. Yeast extract supplies vitamins and
other growth factors. Agar is added as a solidifying agent. Glucose is
a carbohydrate source and acts as a reducing agent. Haemin is
required by a variety of obligate anaerobes. Tris buffer prevents an
extreme drop in pH during glucose fermentation. Cysteine acts as a
reducing agent and stimulates the growth of some anaerobes. Vitamin
K is a growth requirement for some strains of Prevotella
melaninogenica and enhances the growth of some Bacteroides
fragilis and Gram-positive non-spore formers. Sheep blood supplies
haemin and additional growth promoting substances.

Storage
This product is ready to use and no further preparation is necessary.
Store product in its original packaging at 2‒12°C until used.
Store away from light.
Allow product to equilibrate to room temperature before use. Do not
incubate prior to use.
Specimen Collection, Handling and Storage
Specimens should be collected and handled following the
recommended guidelines.

Procedure
Refer to any local protocols and guidelines.
(1) Inoculate and streak the specimen onto the medium.
(2) If material is being cultured directly from a swab, roll the swab
over a small area surface and streak for isolation of single
colonies.
(3) Immediately incubate plates anaerobically at 36°C ± 1°C for 48
hours. Plates can be examined for growth after 24 hours.
Prolonged incubation may be required for some anaerobes.

This medium can be tested with the following strains:

It is the responsibility of the user to perform Quality Control testing
taking into account the intended use of the medium, and in
accordance with any local applicable regulations (frequency, number
of strains, incubation temperature etc.).
The product should not be used if:
(1) The product is contaminated
(2) The colour has changed
(3) The expiration date has passed
(4) There are other signs of deterioration
Performance
Performance was evaluated using 18 bacterial strains including the
following: Clostridium spp., Bacteroides spp., Peptostreptococcus
spp., Prevotella spp., Staphylococcus aureus and Enterococcus
faecalis.
All organisms gave expected growth characteristics
according to the current product specification.
