    The 9-way D Type plug on the Kryo550-16 interface panel is only for connecting to the RS232 serial port of a computer which complies with IEC950.

    Chapter 1 Introduction to the Kryo550-16 Welcome to the Kryo550-16, a fully featured tissue bank freezer for the cryopreservation of bone marrow, stem cells, skin, cord blood and other critical high volume samples. This manual is intended for both operators of the freezer and those persons responsible for its installation.

    All results from the run are stored on the PC for traceability purposes. Any profile you run from the PC may be selected to be stored in the Kryo550-16's memory (overwriting any previously stored profile). This enables you to rerun the profile directly from the freezer without having the PC connected.

    Kryo550-16 Profile Memory The Kryo550-16 has two memory locations that are used for storing profiles. These are the Permanent Profile and Run Profile areas. When the Run button on the control panel is used to start a profile, the freezing profile stored in the Permanent Profile memory is copied into the Run Profile location and then started.

    If you do not have the kryo-connect option and want to interface with a PC, connect RS232 serial port connector ( 9 way D type plug) on the side of the Kryo550-16 to an RS232 port on the PC using the cable supplied. This is a straight-through one to one cable.

    The liquid-nitrogen supply pressure must not exceed 2.0 bar (30 psi). If the hose is not already fitted to the Kryo550-16, wrap the male hose thread with PTFE tap and fix to the LN input on the back of the chamber. Tighten securely with a spanner.

    To avoid damage to the heating element, support the pump carefully while it is out of the dewar. If the hose is not already fitted to the Kryo550-16, wrap the male hose thread with PTFE tap and fix to the LN input on the back of the chamber.
  • Page 22 2-4 Installing the Kryo550-16 Fitting the pump Wear protective clothing, including cryogloves, goggles, boots and a protective apron. Ensure that the red pressure-release valve on the pump is open. Ensure that power is disconnected from the pump. Ensure the delivery hose from the pump to the chamber is securely tightened.
  • Page 23 Installing the Kryo550-16 2-5 Testing the Dewar Wear protective clothing, including cryogloves, goggles, boots and a protective apron. Press the switch on the pump power supply unit. The adjacent light will illuminate. After about 5 minutes, the pressure will rise to 5 psi and the light will switch off. The dewar should remain pressurised until almost empty.

    It is normal to load the samples after the chamber has reached the start temperature. If you intend to run from a PC, initiate the profile from the PC using the Planer plc application - DeltaT. Refer to the DeltaT documentation for details.

    Run button on the control panel. This will cause the Kryo550-16 to start heating to its start temperature. Once the Kryo550-16 starts beeping to indicate that the start temperature has been reached, the Stop button can be pressed to end the run.

    When the seeding start temperature is reached, the chamber temperature is maintained at that temperature for the specified soak time. After this period, the Kryo550-16 displays S and beeps. The seed temperature is still maintained, but the operator will normally momentarily remove the samples and seed manually;...
  • Page 28 3-4 Freezing Samples Pulse seeding Long pulse durations can cause the chamber to overcool so this feature should be used with care. When determining the appropriate time always start with a very short duration, less than 5 seconds, and then experiment with longer times until an appropriate duration has been found.

    When you have removed the samples, press the reset button on interface panel . To deal with a system failure Switch off the power to the Kryo550-16 using the mains on/off switch at the rear. Reset the Kryo550-16’s microprocessor. The reset button is operated by inserting the tip of a ball-point pen or similar tool through the hole in the interface panel located just above the manual override buttons.

    Check all mains cables and interconnecting cables for signs of damage The Kryo550-16 is classified as Class I equipment and must be earthed for safe operation The freezer, pump and the mains connecting cords should be regularly checked by suitably trained personnel, using a Portable Appliance Tester or similar equipment, to ensure adequate earth bonding.

    4-2 Routine Maintenance and Troubleshooting Troubleshooting Refer to the safety instructions at the front of this manual. The Chamber will not Cool Check the nitrogen supply. Check that the pump filter is not blocked by ice (if using an LNP4 pump). The Chamber will not Heat Check that the freezer did not overheat (wait until the thermal cut-out resets).
