This instruction manual is for the Olympus Disk scanning unit for IX3 model IX3-DSU.
To ensure the safety, obtain optimum performance and to familiarize yourself fully with the use of the
disk scanning unit for IX3, we recommend that you study this manual thoroughly before operating
the microscope. In addition, be also sure to read the instruction manual for the microscope on which
the disk scanning unit for IX3 is installed.
Retain this instruction manual in an easily accessible place near the work desk for future reference.

    IX3-DSU IMPORTANT CAUTIONS This manual – part or whole, shall not be used or reproduced without permission. Releasing the Transport Lock Be sure to work on the following tasks first after finishing the unpacking. When powering up the control box without removing the CAUTION transportation lock knob, the disk unit may be damaged.

If no power cord is provided, please select the proper power cord by referring to the section “PROPER SELECTION OF THE POWER CORD” at the end of this instruction manual.     · Do not use the instruments where they are subjected to direct sunlight, high temperature and humidity, dust or vibrations. · The fluorescent observation by vision is not available with IX3-DSU, If you perform the observation by vision, use the IX3 system for fluorescent observation separately.

    IX3-DSU Cleanup and storage · Never put dirt or fingerprint on the surface of lens and filter, etc. In case of contamination, use gauze and wipe out lightly. In case that the dirt or fingerprint does not come off with gauze, then, use mixed liquid (alcohol 3: ether 7) to put in gauze and wipe out gently.

    IX3-DSU SYSTEM OUTLINE Attaching this system to a microscope, it comprises a confocal microscope system without laser, but with use of a white light source. It acquires a high contrast image through resolution enhanced in light axis direction very rapidly.

    SUMMARY OF OBSERVATION PROCEDURES This section describes the procedures for performing the fluorescent observation with vision by using your reflected fluorescent system and the observation with camera and the confocal observation by using the DSU system continuously. For the fluorescent observation with vision, refer to the instruction manual of [Reflected Fluorescent System] provided separately. Also, for operations of the microscope IX83, refer to the instruction manual of [IX83] together with the above.

    3-7 Selecting Mirror Unit built-in DSU Press the button of the hand switch to rotate the mirror unit, and engage the desired mirror unit into the light path. In the factory default setting, the button to rotate the mirror unit is assigned to the buttons (1) and (2) shown in the picture on the left.

    ADJUSTMENT 8-1 Centering the Field Stop } Use the Allen screwdriver provided with the microscope to operate the centering screws. Close the shutter of the manual shutter. (See (1) in the picture.) Engage the fluorescent mirror unit of B excitation into the light path. Open the shutter of the mirror unit cassette.

    IX3-DSU 8-2 Centering of the Lamp } It is not necessary to perform the centering of the lamp in each observation, but perform the centering when replacing the lamp or if you recognize the uneven brightness in the acquired image.

    IX3-DSU 9-2 Attaching the DSU adapter Loosen the clamping screw (1) of the DSU adapter by using the Allen screwdriver provided with the microscope frame, and detach the DSU adapter (2) from the disk box. Loosen the side port clamping screw (3) placed on the left side surface of the microscope frame by using the Allen screwdriver provided with the microscope frame.

Loosen the clamping screw (3) (2 positions) of the disk box by using the Allen screwdriver provided with the microscope frame.

SHI-130 OL) Before replacing the lamp, be sure to turn \ (OFF) the power CAUTION switch and unplug the power cord from the outlet, and wait (more than 40 minutes as a guide) until the lamp replacement cover and the lamp are cooled down sufficiently.

    Marcury lamp } If your system uses the xenon burner, attach the burner, set the hour counter and connect the system by referring to the instruction manuals for the lamp housing and power supply unit. Loosen the socket clamping screw using the Allen screwdriver. Hold the upper section of the lamp housing and pull it upward to remove the socket section.

    9-11 Disk installation and change To replace the disk, turn off the mainswitch of the control box, CAUTION and wait 1 minute until the disk stops rotating. Loosen screws (2) (2 places) at front side of access panel (1) located on top of the disk box, using the Allen screwdriver and remove the access panel.

    9-14 Attaching the spacer ◎ For IR observation (when using GFP cubes), be sure to attach the spacer provided with IX3-DSU in order to secure the optical perfor- mance. Loosen the fixing screws (1) (2 places) of the filter wheel by using the Allen screwdriver provided with the microscope.

    9-15 Field iris diaphragm IN/OUT (Removal). Loosen fixing screw (1) located at the bottom of aperture unit, using the Allen screwdriver. Hold aperture unit (2) and pull it horizontally. (Installation). Reverse procedure When you tighten fixing screw (1), tighten it lightly. CAUTION Do not tighten it too hard.

    IX3-DSU 9-17 Attaching the Camera Loosen fixing screw (1) of port, using the Allen screwdriver that comes as accessory for microscope frame and remove C-mount adapter (2) inside the port. Screw in and fix the C-mount adapter (2) that was removed to C-mount section (3) of camera firmly.

    9-18 Attaching the Motor Drive Board You must install motor drive board after you turn power of CAUTION control box to OFF. Loosen the four fixing knobs for the two option slot covers at the left side of the back panel of the control box (1) and remove the covers and knobs.
